Eluxadoline is a prescription medication used to manage irritable bowel syndrome with diarrhea (IBS-D) in adults. It works locally in the gut as a mixed opioid receptor agonist and antagonist, helping to reduce bowel contractions and improve symptoms like diarrhea and abdominal discomfort.

Indications

Irritable Bowel Syndrome with Diarrhea (IBS-D) in adults.


Dosage

- Recommended dose: 100 mg orally twice daily with food. - For patients without a gallbladder or with mild liver impairment: 75 mg twice daily. - Should be taken exactly as prescribed by the physician.


Contra-Indications

- Known hypersensitivity to Eluxadoline. - Severe liver impairment. - Biliary duct obstruction or history of pancreatitis. - History of alcoholism or heavy alcohol use. - Severe constipation or gastrointestinal obstruction.


Special Precautions

- Use with caution in patients with mild to moderate liver impairment. - Not recommended for patients with a history of pancreatitis or alcoholism. - Monitor for signs of severe constipation or abdominal pain. - Avoid use with medications that slow gastrointestinal motility.


Side Effects

Common side effects may include: - Nausea - Abdominal pain or discomfort - Constipation - Vomiting - Dizziness Serious side effects (require immediate medical attention): - Severe abdominal pain - Signs of pancreatitis (persistent upper abdominal pain, nausea, vomiting) - Allergic reactions (rash, itching, swelling)


Drug Interactions

- Avoid strong CYP3A4 inhibitors (e.g., ketoconazole, clarithromycin). - Use caution with other opioids or medications that cause constipation. - Concomitant use with eluxadoline and alcohol may increase risk of liver injury.
