Lomustine (Nitrosoureas)

Lomustine is an oral alkylating agent from the nitrosourea class, used primarily as an antineoplastic (anticancer) medication. It works by interfering with DNA and RNA synthesis, leading to cancer cell death. Lomustine is known for its ability to cross the blood–brain barrier, making it particularly valuable in treating brain tumors. It is highly lipid soluble alkylating agent with a wide range of antitumour activity. They cross blood brain barrier and are effective in meningeal leukemias & brain tumours.

Indications

Meninigeal leukemias, brain tumours, Hodgkin's disease.


Dosage

100-130mg/sqm body surface area single oral dose every 6 wks.


Contra-Indications

Pregnancy, lactation, Hypersensitivity.


Special Precautions

Renal impairment. C.N.S disturbances, Monitor blood counts. Paediatrics: Not recommended in infants. Pregnancy : Safety not established. Lactation : Safety not established. Elderly : Use with caution.


Side Effects

Nausea, vomiting, seizures, Bone marrow depression is delayed for 6 wks. Visceral fibrosis and renal damage.


Drug Interactions

Theophylline cause leucopenia and thrombocytopenia.
